The Crz1-like protein FolSrz1 contributes to oxidative stress resistance and virulence through maintenance of cellular homeostasis in Fusarium oxysporum f. sp. lycopersici
Listed in NCBI GEO
The zinc finger transcription factor Crz1 is a conserved downstream target of the Ca2+-calcineurin signaling pathway in many fungi.
Description
However, its role in the tomato wilt fungus Fusarium oxysporum f. sp. lycopersici (Fol) remains unclear. In this study, we identified and functionally characterized FolCrz1, an ortholog of yeast Crz1, and FolSrz1 (stress resistance zinc finger 1), a C2H2 zinc finger protein that shares partial sequence identity with F. graminearum FgCrz1A.
The ΔFolcrz1 mutant displayed increased sensitivity to Ca2+, and GFP-fused FolCrz1 translocated to the nucleus in a Ca2+/calcineurin-dependent manner. However, deletion of FolCRZ1 did not affect virulence. In contrast, the ΔFolsrz1 mutant showed impaired growth, increased sensitivity to oxidative and membrane stress, and markedly reduced virulence.

Deletion of FolSRZ1 also reduced fungal biomass in tomato roots during infection. FolSrz1 was partially localized to the nucleus under basal conditions. Transcriptomic analysis further revealed that FolSrz1 primarily affects the expression of genes associated with cellular homeostasis, including genome integrity, proteostasis, and copper homeostasis.
Our findings identify FolSrz1 as a previously uncharacterized Crz1-like transcription factor that promotes fungal fitness by regulating the basal expression of genes involved in cellular homeostasis, thereby contributing to oxidative stress resistance and virulence.
