Gongzhu Cuo Modern Sample GDGTs Compound Dataset

Listed in ScienceDB

This dataset contains 27 modern samples from the Gongzhucuo watershed in the southwest of the Qinghai Tibet Plateau, including 17 watershed surface soil samples, 9 lake surface sediment samples, and 1 river surface sediment sample.

Description

All samples were subjected to GDGTs compound analysis, and their pretreatment and computer analysis were completed in the laboratories of the China University of Geosciences (Wuhan) and the Qinghai Tibet Plateau Research Institute of the Chinese Academy of Sciences.

After freeze-drying, modern samples are thoroughly ground and sieved to remove impurities such as plant roots and stones, and finally ground into a uniform powder. Weigh about 5 g of the sample and perform ultrasonic extraction using a mixed solvent of dichloromethane and methanol (dichloromethane: methanol=9:1, V/V). Vortex the mixture and place the centrifuge tube symmetrically in the centrifuge for centrifugation.

The supernatant after centrifugation is concentrated by rotary evaporation and transferred to the corresponding container, and then dried with nitrogen gas. After saponification treatment and concentration, the obtained extract was further separated by silica gel column chromatography, using n-hexane and dichloromethane to separate non-polar and polar components. Collect polar components and blow dry them, then dissolve them again using a mixture of n-hexane and isopropanol solvent (99:1, V/V), filter through a 0.45 μ m hydrophobic membrane, and use the resulting sample for subsequent instrument analysis.

The pretreatment of all modern samples was completed in the State Key Laboratory of Biogeology and Environmental Geology of China University of Geosciences (Wuhan). GDGTs analysis of modern samples was completed in the State Key Laboratory of Biogeology and Environmental Geology of China University of Geosciences (Wuhan) and the Qinghai Tibet Plateau Research Institute of the Chinese Academy of Sciences. Among them, China University of Geosciences (Wuhan) used Agilent 1200 series high-performance liquid chromatography combined with Agilent 6460A triple quadrupole mass spectrometer (HPLC-MS/MS) for semi quantitative determination of GDGTs in the sample; The Qinghai Tibet Plateau Research Institute of the Chinese Academy of Sciences uses a high performance liquid chromatography atmospheric pressure chemical ionization mass spectrometer (Agilent 1260 HPLC – 6100 MS) equipped with an automatic sampler to analyze and determine GDGTs.
