Intact Cell Dissociation of FFPE Colon Tissue Enhances Cell Recovery and Gene Detection in scRNA-seq

Formalin-fixed, paraffin-embedded (FFPE) tissues present challenges for transcriptomic profiling due to RNA crosslinking and fragmentation.

Description

The 10x Genomics Chromium Fixed RNA Profiling (Flex) platform is designed for single-nucleus profiling of FFPE tissue. Here, we extend this workflow by implementing a whole-cell dissociation approach for FFPE colorectal tissue to improve epithelial cell recovery and compare its performance with the standard single-nucleus dissociation protocol.
