Multifunctional nanoparticles for photodynamic and hypoxia-activated cancer therapy

Photodynamic therapy (PDT) is a therapeutic method corresponding to excitation of a molecule, the photosensitizer, by near-infrared (NIR) light.
Description
The excited photosensitizer reacts with oxygen which turns into reactive oxygen species (ROS) and then might kill cells like, for example, tumorous cells. PDT is attractive as it may enable for achieving a non-invasive therapeutic action.
Nevertheless, PDT is limited by the absence of oxygen. In a medium for instance, if all the oxygen is consumed, PDT can no longer be implemented. The hypoxia-activated prodrug Cu(TPZ)2 (TPZ: tirapazamine) was studied in our team.

The main objective of this research project is to combine the photosensitizer indocyanine green (ICG) and Cu(TPZ)2 in a single liposomal formulation to circumvent the oxygen-related limit of PDT mediated by ICG. In this project, the two main liposomal formulations used were DOPC:chol:DSPE-PEG (0.95:0.5:0.05) and DSPC:chol:DSPE-PEG (0.95:0.5:0.05). ICG-loaded liposomes were produced by implementing bilayer loading method, passive loading method, or remote loading method.
Moreover, Cu(TPZ)2-loaded liposomes were also produced using the same loading methods. The bilayer loading method and the remote loading method for ICG and Cu(TPZ)2, respectively, were selected to produce ICG/Cu(TPZ)2-co-loaded liposomes. This work set up the basis for further in vitro studies.<br>
