Single-Nucleus Multiome Analysis of Atherosclerotic Arteries from Prdm16os KO and Control Mice

Listed in NCBI GEO

Through comprehensive omics data mining, we identified that the lncRNA PRDM16-DT (mouse ortholog: Prdm16os) is enriched in contractile vascular smooth muscle cells (VSMCs) and downregulated during VSMC phenotypic switching in atherosclerosis.

Description

PRDM16-DT is divergently transcribed from PRDM16 gene within a coronary artery disease (CAD) GWAS locus. Fine-mapping and colocalization analyses revealed that PRDM16-DT is the most likely causal gene underlying this association signal.

Global deletion of Prdm16os accelerates atherosclerosis in mice. To further understand the underlying molecular mechanisms, we performed single-nucleus RNA and chromatin accessibility profiling using the 10x Genomics sginel cell multiome ATAC + RNA platform on atherosclerotic aortas from Prdm16os KO and control mice.
