Description
Based on the published genome sequence of D. officinale,SSR loci were identified using MISA software, and SSR molecular markers were designed via Primer 3.Genomic DNA of D. officinale was extracted and subsequently detected via gel electrophoresis following the protocol described by Pan et al..
To develop genome-wide SSR markers, the published D. officinale genome (NCBI assembly accession: ASM160598v2, GCA_001605985.2. The website is ncbi.nlm.nih.gov/datasets/genome/GCF_001605985.2/) was utilized. The MISA (MicroSatellite identification tool; pgrc.ipk-gatersleben.de/misa/) software was employed to screen for polymorphic microsatellite loci across the genome.

High-throughput primer design was performed using Primer 3 (primer3.ut.ee/). The design parameters were as follows: primer length of 18–25 bp, expected PCR product size of 100–250 bp, and GC content of 40–60%. The Perl encoded text has been uploaded to the database
