In vivo-based CRISPR screen identifies ZC3H12C as a mediator of CAR-T cell dysfunction in solid tumors

Listed in NCBI GEO

In this work, we have sought to develop an in vivo model of xenograft tumors that recapitulates disease recurrence in absence of antigen modulation, to study the T cell-intrinsic mechanisms that give rise to CAR-T cell dysfunction.

Description

We have developed an in vivo model in which CAR-T cell become dysfunctional. We isolated tumor infiltrating CAR-T cells after long exposure to tumors and studied the transcriptome.

Using this newly generated knowledge, we have identified gene candidates whose genetic deletion could enhance CAR-T antitumor effect in a model of solid tumors.
