Generation and trial of mucosal virus vectored vaccines for bovine respiratory viruses

Bovine respiratory syncytial virus (bRSV) and bovine parainfluenza virus type 3 (bPIV3) are important components of the bovine respiratory disease complex (BRDC), causing significant bovine morbidity and mortality.
Description
Current vaccine strategies arein efficient, and there is a clear need for novel vaccine candidates which subvert maternal antibody interference. Replication competent (rc) and replication incompetent (ri) Sendai virus (SeV) vectored vaccines expressing bRSV and bPIV3 surface glycoproteins were generated from their corresponding cDNA clone plasmids and characterised in vitro to confirm vector and heterologous protein expression.
(rc) and (ri) vectored vaccines expressing bRSV heterologous proteins were taken forward for an immunogenicity study in mice, where bRSV specific IgG and mucosal IgA, as well as virus neutralisation capability, were investigated. Several promising vaccine candidates were identified which induced high levels of bRSV specific IgG and IgA, and significantly higher virus neutralisation antibody titres than controls.<br><br><i>Thesis is embargoed until 31 July 2028.</i>
