Genetically modifying the RNA-binding protein Regnase-1 reveals its critical role in Treg homeostasis and functions in vivo [HyperTRIBE]

Listed in NCBI GEO

In HyperTRIBE analysis, a hyperactive mutant of the catalytic domain of ADAR enzyme is fused to an RNA-binding protein of interest in order to identify RNAs that are bound by the protein interest through RNA-sequencing (Rahman et al Nat Protoc. 2018).

Description

By discovery of RNAs where adenosines have been edited to iosines, the RNAs that were bound by the protein of interest and then edited by ADAR are identified genome-wide.

We conducted this analyis for Regnase-1, an RNA binding protein with multiple described cellular functions, including mRNA degradation and T cell activation. Because Regnase-1 can degrade the mRNA quickly, we used the nuclease-dead mutant Reg1-D141N to conduct this assay in activated CD4+ T cells from wild-type mice.
