Nonsense-mediated decay mRNA quality control system is essential for root development and efficient root nodule symbiosis in Medicago truncatula
Listed in NCBI GEO
Quality control systems ensure the fidelity of gene expression by identifying and eliminating aberrant gene products.
Description
Nonsense-mediated decay (NMD) mRNA surveillance pathway recognizes and degrades aberrant mRNAs containing premature termination codons. Additionally, NMD regulates the expression of several normal genes, and in Arabidopsis, it also modulates antibacterial defense by controlling the expression of resistance genes.
Surprisingly, the role of quality control systems in root nodule symbiosis has remained unexplored. To investigate the function of NMD in nitrogen-fixing symbiosis, we attempted to inactivate the core NMD factors UPF1 and UPF2 in Medicago truncatula. Since the complete knockout of these genes was not achievable, we conclude that NMD is essential for root growth.

We therefore generated upf1 and upf2 NMD-deficient RNAi roots and inoculated them with Sinorhizobium medica. Both infection events and nodule formation were significantly reduced in the RNAi roots, indicating that NMD is required for efficient symbiosis; even partial inactivation of NMD impairs infection. In line with the sensitivity of symbiosis to NMD intensity, we identified multiple autoregulatory circuits that stabilize NMD activity in M. truncatula.
Finally, RNA-seq revealed pronounced alterations in symbiosis-related pathways, including flavonoid biosynthesis and defense responses, in NMD-deficient roots, likely explaining the defects in rhizobia infection.
