Dataset of soil extracellular enzyme activities, enzyme stoichiometric ratios, and soil physicochemical properties from Xianshan, Xinyang, Henan, China (August 2026)
Listed in ScienceDB
This dataset is derived from soil sample measurements obtained from long-term nitrogen addition and drought treatment experiments.
Description
After soil sample collection, it is used to determine soil extracellular enzyme activity and soil physicochemical properties. Extracellular enzyme activity indicators include cellulase (CBH), β - glucosidase (β G), acid phosphatase (AP), and β - N-acetylglucosamine (NAG); The indicators of soil physical and chemical properties include soil pH, soil moisture content (SWC), microbial biomass carbon (MBC), dissolved organic carbon (DOC), nitrate nitrogen (NO3-- N), and ammonium nitrogen (NH4+- N). In the process of data processing, first, sample numbering, processing grouping, and unit organization are performed on the measured enzyme activity and soil physicochemical properties data; Subsequently, enzyme stoichiometry indicators EC: N, EC: P, EN: P, as well as vector angle (Vector A) and vector length (Vector L), were calculated based on CBH, β G, AP, and NAG enzyme activity data.
The dataset consists of the organized mean and standard deviation. The soil pH was measured using a glass electrode pH meter (STARTER300, OHAUS, USA) with a soil water mass ratio of 1:2.5; Moisture content is determined by drying method; Soil microbial biomass carbon (MBC) was measured using chloroform fumigation method; Soluble organic carbon (DOC) was extracted by KCL solution and measured by a total organic carbon analyzer (TOC-V CPH analyzer, Shimadzu Corporation, Japan); Nitrate nitrogen (NO3-- N) and ammonium nitrogen (NH4+- N) were extracted using KCL solution and subsequently measured using a continuous flow analyzer (Skalar San++, Netherlands). This experiment measured the activity of four extracellular enzymes in soil, including cellulose hydrolase (CBH), β - glucosidase (β G), acid phosphatase (AP), and β - N-acetylglucosamine (NAG).

The substrates used are 4-MUB-beta-D-glucoside, 4-MUB-beta-D-glucoside, 4-MUB-phosphate, and 4-MUB-N-acetyl-D-glucosamine. After adding the substrate, the sample was incubated at 25 ° C in the dark for 4 hours, and the fluorescence intensity was detected using a multifunctional enzyme-linked immunosorbent assay (SpectraMaxM5, Molecular Devices, USA).
