Description
This dataset is the raw data, processing data and visualization files of all experimental characterization and biological evaluation in the process of constructing polydopamine nanoparticles (PDA NPs) composite ROS-responsive hydrogel (Gel), which completely records the whole process data from material preparation, physical and chemical characterization to cellular/molecular level functional verification. The dataset was divided into six groups according to the thesis chapters, covering the key experiments such as material characterization, in vitro antioxidant, rheology, cytocompatibility, ROS scavenging, apoptosis and immune regulation.
All experiments were completed between November 2024 and April 2026, and no data were missing.I. Data set generation process and processing method1. Data related to nanoparticles (PDA NPs)Content: The nanoparticles were characterized by their morphology, size distribution, Zeta potential, and ROS scavenging ability (hydroxyl radical scavenging, H₂O₂ scavenging, superoxide anion scavenging). The production process: PDA NPs were synthesized by oxidation self-polymerization; Scanning electron microscope (SEM) and transmission electron microscope (TEM) were used to observe the microstructure.

Particle size distribution and Zeta potential were measured by dynamic light scattering (DLS). ROS scavenging related indicators were measured by kit method/spectrophotometry. The absorbance data were recorded and the clearance rate was calculated.
Methods
Excel was used to sort out the original absorbance data and calculate the clearance rate. After the particle size distribution and Zeta potential data were exported using the built-in software of the instrument, the statistics were sorted out in Excel, and finally GraphPad Prism was used to generate charts and export as EMF format vector charts.2. Hydrogel (PDA/Gel) related dataContent: The macroscopic morphology, microstructure, rheological properties (viscosity curve, energy storage/loss modulus), ROS responsiveness, and antioxidant activity (DPPH method) of the hydrogel were characterized.TSPBA-PVA cross-linked hydrogel was prepared and loaded with PDA NPs to obtain PDA/Gel.
The internal microstructure was observed by SEM. Viscosity scanning and oscillation scanning were performed using a rheometer to record rheological data. ROS responsiveness The antioxidant activity of the hydrogel extract was measured by the DPPH assay, and absorbance data were recorded.
Methods
Rheological data were exported by the rheometer software, sorted in Excel, and drawn viscosity curve and modulus curve by GraphPad Prism. The DPPH experimental data were used to calculate the clearance using Excel, and finally all charts were exported as EMF format vector maps, and SEM images were directly saved as original images.3. Cytocompatibility and antioxidant activity data (HT22 cells)Content: The cytotoxicity of PDA NPs and PDA/Gel extract on HT22 cells was characterized by CCK-8 method and live/dead cell staining.Production process: HT22 cells were co-cultured with different concentrations of PDA NPs or PDA/Gel extract.
CCK-8 reagent was used to detect cell viability and the absorbance value was recorded. After Calcein-AM/PI staining, live/dead cell images were taken by fluorescence microscopy.
Methods
CCK-8 data were analyzed by Excel to calculate cell viability, GraphPad Prism was used to draw bar charts and exported to EMF format. Live/dead staining images were sorted by group and saved as original images without missing data.4. Intracellular ROS scavenging and apoptosis data (HT22 cells)Experimental content: DHE staining was used to detect intracellular ROS level, and TUNEL staining was used to detect cell apoptosis. The production process: HT22 cells were co-cultured with PDA/Gel extract after OGD/R model was established.
After DHE staining, ROS-positive cell images were taken by fluorescence microscope. After TUNEL staining, images of apoptotic cells were taken by confocal microscopy.Processing methods: All fluorescence images were sorted according to groups and saved as original images without missing data.5. Microglial polarization and inflammatory factor expression data (BV2 cells)Experimental content: CD16/32 (M1) and CD206 (M2) immunofluorescence staining, and qPCR was used to detect the mRNA expression levels of inflammatory factors (IL-1β, IL-6, and TNF-α).Production process: BV2 cells were polarized by LPS and co-cultured with PDA/Gel extract.
CD16/32 and CD206 positive cell images were taken after immunofluorescence staining. Cellular RNA was extracted, and the mRNA expression of inflammatory factors was detected by qPCR after reverse transcription, and the Ct value data were recorded. Methods: qPCR data were analyzed by 2^-ΔΔCt method in Excel to calculate the relative expression of QPCR data, GraphPad Prism was used to draw bar charts and exported to EMF format.
Immunofluorescence images were organized by group and saved as original images without missing data.6. Microglial and neuronal co-culture data (HT22 cells)Experimental contents: live/dead staining and TUNEL stainingProduction process: Live/dead staining and TUNEL staining were performed on HT22 cells after co-culture with BV-2 cells treated in different waysProcessing methods: All fluorescence images were sorted according to groups and saved as original images without missing data.Ii.
Data file and format descriptionFile format and applicable toolsExcel file (.xlsx) : Contains raw data, statistics, and calculation results for all experiments, and can be imported and processed using Microsoft Excel, WPS Office, or GraphPad Prism.EMF file (.emf) : Vector chart file exported for GraphPad PrismImage file (in folder) : Contains original images such as SEM, fluorescence microscope and physical photos in JPG/PNG/TIF format, which can be processed using software such as ImageJ and Photoshop.3.
Data error and explanationAll experiments in this dataset were set up with at least three biological replicates. The data error is the standard error of repeated experiments (SEM), which has been reflected in Excel and Graphpad charts. The main sources of error were experimental operation error, instrument detection error and individual differences among biological replicates.
The error range was within the acceptable range and did not affect the reliability of the experimental conclusion.
