Supplemental Material for: Drug persistence and treatment outcomes after switching from intravenous to subcutaneous vedolizumab in inflammatory bowel disease

In this single-center prospective cohort, 51 patients with Crohn’s disease (CD, n=27) or ulcerative colitis (UC, n=24) IV-to-SC transitionedand were followed for 24 months. Patients who declined switching continued IV therapy (n=24) and served as a control group. Clinical indices, fecal calprotectin (FCP), VDZ trough levels, and health-related quality of life (HRQoL) were assessed at baseline, and at 3, 12, and 24 months.

Drug persistence was analyzedusing Kaplan–Meier and Cox regression models. </p><p dir="ltr">Results: SC persistence was 96%, 86%, and 76% at 3, 12, and 24 months, respectively, with no statistically significant difference compared with IV therapy.Clinical indices, biochemical remissionrates, andHRQoL remained stableover all time points following IV-to-SC switch. FCP increased modestly after 12 months in SC-treated CD patients, without a corresponding statistically significant deterioration in clinical indices or biochemical remission rates.VDZ trough levels increased after switchingand were associated with biochemical remission.

Most patients experienced the IV-to-SC switch aspositive. </p><p dir="ltr">Conclusion: Switching from IV-to-SC VDZ wassafe, effective, and well accepted by patients.No statistically significant differences in two-year persistence, clinical, and biochemical outcomes were observed compared with continued IV therapy.SC VDZ is thus a viable, patient-centered maintenance option for IBD.</p>
