RNA-seq Profiling of shB7H3 and Scramble E0771 Tumors in Control and Diet-Induced Obese Mice

Listed in NCBI GEO

Obesity promotes triple-negative breast cancer (TNBC) progression in part through immunosuppressive remodeling of the tumor microenvironment.

Description

B7-H3 (CD276), a co-inhibitory immune checkpoint molecule, is frequently overexpressed in TNBC and is regulated by obesity-associated inflammatory cytokines. To define the transcriptional consequences of B7-H3 knockdown in TNBC tumors in lean and obese mice, we performed bulk RNA-seq on whole E0771 mammary tumors engineered to stably express either a B7-H3-targeting shRNA (shB7H3) or a scramble shRNA control.

Tumors were harvested from orthotopically engrafted C57BL/6 female mice maintained on a control (10% kcal fat) or high-fat diet-induced obese (60% kcal fat) protocol. Transcriptomic analysis and GSEA revealed that B7-H3 suppression in obese mice promoted inflammatory response, interferon signaling, and IL6/JAK/STAT3 pathway signatures.
