Metabolomic dataset of subcellular uranium partitioning and metabolic reprogramming in rice roots modulated by transport inhibitors

Listed in ScienceDB

Description

This dataset was generated from a hydroponic experiment investigating the metabolic responses of rice (Oryza sativa L.) roots to uranium (U) stress, as well as the modulatory effects of transport regulators (AgNO₃ as an aquaporin modulator and glycerol as an osmotic regulator) on U uptake, toxicity, and subcellular distribution. Nine treatment groups were established, including control, U alone (20 µM and 200 µM), AgNO₃ alone, glycerol alone, and combined treatments of U with AgNO₃ or glycerol.

Root samples were collected 24 h post-treatment and subjected to untargeted metabolomics profiling using ultra-high performance liquid chromatography-tandem mass spectrometry (UHPLC-MS/MS).The dataset contains metabolite information detected in both positive and negative ionization modes, including raw MS data, pre-processed peak area matrices, metabolite identifications (based on matching against public databases including mzCloud, MassBank, METLIN, and an in-house library), and differentially accumulated metabolite screening results (VIP ≥ 1, |log₂FC| ≥ 1, p < 0.05).

Data quality and group separation were validated by principal component analysis (PCA) and orthogonal partial least squares discriminant analysis (OPLS-DA).Key findings from this dataset reveal that U treatment alone induced 55 differential metabolites (37 upregulated and 18 downregulated), primarily affecting starch and sucrose metabolism and pantothenate and CoA biosynthesis. AgNO₃ co-treatment triggered 199 differential metabolites (16 upregulated and 183 downregulated), broadly suppressing pathways related to amino acid metabolism, ABC transporters, and aminoacyl-tRNA biosynthesis.

Glycerol co-treatment resulted in 93 differential metabolites (21 upregulated and 72 downregulated), with upregulation of TCA cycle intermediates and ether lipid metabolism alongside suppression of amino acid and nucleotide metabolism, reflecting a metabolic trade-off prioritizing energy production over biosynthesis. This dataset provides fundamental data supporting research on phytoremediation of U-contaminated farmlands, safe rice production, and the regulatory mechanisms of metabolic acclimation to heavy metal stress.
