Transcriptomic analysis of perfluorobutane Sulfonic Acid (PFBS) in two human placental models

Listed in NCBI GEO

Perfluorobutane sulfonic acid (PFBS) is a short-chain replacement per- and polyfluoroalkyl substance (PFAS) increasingly detected in drinking water and human serum.

Description

Prenatal PFBS exposure has been epidemiologically associated with hypertensive disorders of pregnancy, including preeclampsia, and increased offspring adiposity; however, the toxicological mechanisms underlying these associations and the effects of PFBS on human placental tissue remain poorly characterized.

With a transplacental transfer efficiency of approximately 97%, PFBS undergoes near-complete transfer to the fetal compartment, underscoring the placenta as a critical target for PFBS toxicity. This study investigated the transcriptional and functional effects of PFBS in human trophoblast cells and term placental villous explants, stratified by fetal sex, to characterize mechanisms of PFBS bioactivity relevant to placental function.

Term chorionic villous explants (obtained from with informed consent from elective non-laboring c-sections) from male and female placentas were exposed to 5 or 20 µM PFBS and analyzed by RNA-seq with sex-stratified differential gene expression and gene set enrichment analysis (GSEA) using the MSigDB Hallmark gene set collection.
