BASIC PENTACYSTEINE 1 (BPC1) is involved in the epigenetic suppression of camalexin biosynthesis in Arabidopsis

Listed in NCBI GEO

Camalexin is one of indolic phytoalexins protecting plants against biotic challenges like pathogen attack.

Description

However, molecular mechanism on how camalexin accumulation is modulated is not fully understood. In our study, we found that a plant specific transcription factor, BASIC PENTACYSTEINE 1 (BPC1) play a negative role in the camalexin biosynthesis.

The bpc1-1 mutant has more accumulated amount of camalexin than level of Col-0. In a consistency, expression of genes involved in the camalexin biosynthesis was upregulated in the bpc1-1 mutant in comparison to Col-0. ChIP-qPCR analysis showed that BPC1 directly binds to promoter region of MYB51, CYP79B2, and CYP79B3 via the recognition of GA-rich motif.

In addition, we found that BPC1 is required for the PRC2-medidated enrichment of H3K27me3 on many of camalexin biosynthetic genes. Amount of camalexin was significantly increased in knock-out mutants of components of PRC2 complex like CLF, SWN, and EMF1. Overall, our study demonstrated that BPC1 is required for the epigenetic suppression of camalexin biosynthesis by coordinating the PRC2-mediated enrichment of H3K27me3 in Arabidopsis.
