Dataset of soil environmental factors and bacterial community characteristics in wetlands under different vegetation restoration modes after Spartina alterniflora removal
Listed in ScienceDB
The research area is located in the Minjiang River estuary wetland in Fuzhou City, Fujian Province (25 ° 50 ′ 56 ″ -26 ° 12 ′ 42 ″ N, 119 ° 16 ′ 30 ″ -119 ° 42 ′ 45 ″ E).
Description
Field sampling was conducted in January 2024, and five habitat types were set up for the study: Phragmites australis wetland restored to salt marsh vegetation (T1, 9 sampling points) and Cyperus malaccensis wetland restored to salt marsh vegetation (T2, 8 sampling points), and Kandelia obovata mangrove restoration area artificially planted with autumn eggplant, including 6-year-old (T3, 4 sampling points) and 16-year-old (T4, 8 sampling points).
Two forest age gradients (4 sampling points) and a control group (CK, 3 sampling points) without restoration after treatment, totaling 28 sampling points. The determination of soil physical and chemical properties is carried out using the following methods and equipment: pH (water to soil ratio 1:5) is measured using an IQ Instruments portable pH meter (USA); The conductivity (EC) and soil temperature (ST) were measured using a portable conductivity meter (Spectrum, USA) model 2265FS; The moisture content (SM) is determined by drying method, and the bulk density (BD) is determined by ring cutting method; After leaching with 2 mol · L ⁻¹ KCl solution, ammonium nitrogen (NH ₄⁺ - N) and nitrate nitrogen (NO ∝⁻⁻ - N) were quantitatively detected using a Skalar Analytical San++continuous flow analyzer (Netherlands); Organic matter (SOM) was determined using the loss on ignition method; Total carbon (TC) and total nitrogen (TN) were measured using an Elementar Vario MAX elemental analyzer (Germany); Microbial biomass nitrogen (MBN) was analyzed using chloroform fumigation leaching method.

Soil bacterial community analysis was based on high-throughput sequencing of 16S rRNA genes: total soil DNA was extracted using the OMEGA Soil DNA Kit, and the V3-V4 hypervariable region was amplified using primers 338F (5 '- ACTCCTACGGGAGGCAGCA-3') and 806R (5 '- GACTACHVGGGTWTCTAAT-3'). The PCR products were purified and a library was constructed using Illumina TruSeq Nano DNA LT Library Prep Kit. After evaluating the quality of the library using Agilent Bioanalyzer 2100, PE250 double ended sequencing was performed on the Illumina NovaSeq platform.
After quality control, de chimerism, and de redundancy of the original sequence, the ASV (Amplitude Sequence Variant) algorithm was used for clustering, and seven levels of annotation were completed based on standard taxonomic databases, including boundary, phylum, class, order, family, genus, and species; The ASV abundance matrix is randomly flattened based on 95% of the samples with the lowest sequence size among all samples.
The absolute abundance of bacteria was obtained through 16S rRNA gene absolute quantitative PCR to reflect the bacterial gene copy number per gram of dry soil. The "Soil Physical and Chemical Factors" table consists of 33 rows, including 28 individual samples (T1-1~T1-9, T2-1~T2-8, T3-1~T3-4, T4-1~T4-4, CK-1~CK-3) and 5 mean rows for treatment groups (T1, T2, T3, T4, CK). The "Bacterial Abundance" table consists of 28 rows and records the absolute copy number (copies · g ⁻¹) of the bacterial 16S rRNA gene for each individual sample.
