RNA sequencing of meningeal lymphatic vessels in dural metastasis models

However, recently emerging studies suggest the dura mater is an immune hub and implements immunosurveillance of the central nervous system. These evidences imply potential immunotherapies of DM.

Here, we established a preclinical murine model of DM. We have employed a RNA sequencing approach using SMART-seq to study the meningeal lymphatic vessels of DM model. Briefly, on day 12, the dorsal dura maters were isolated from the head of 4T1-DM mice treated with PBS, ZOL or ZOL+Ang2, and were enzymatically digested to single cells with collagenase VIII and DNase I. The meningeal lymphatic endothelial cells (LECs) were harvested using fluorescent activated cell sorting, CD45-CD31+PDPN+ dural cells were sorted for sequencing.
