TNF-blockade attenuates the proinflammatory CRS signature of monocyte-derived macrophages.

Listed in NCBI GEO

TNF-blockade abates cytokine release, weight loss and eventual lethality in our preclinical model of CAR T cell-induced Cytokine Release Syndrome (CRS).

Description

Macrophages are known mediators in the pathophysiology of CRS. We generated this single-cell RNA sequencing data set to assess the effect of TNF on monocytes and macrophages in CRS.

The data show that upon CRS-induction there is an increase in monocyte-derived (Ccr2-positive) macrophages at the tumor site. In an unperturbed CRS these monocyte-derived macrophages show high levels of Ly6C and accordingly have a proinflammatory phenotype expressing known mediators of CRS-pathophysiology Nos2 and Il1b. Under TNF-blockade the emerging monocyte-derived macrophages show low levels of Ly6C and lack this proinflammatory phenotype.
