Transcriptome profiling of peripheral blood mononuclear cells from patients with alcohol-related hepatitis following antioxidant and trace element supplementation

This study investigated the effects of antioxidant and trace element supplementation on gene expression in peripheral blood mononuclear cells (PBMCs) from patients with alcohol-related hepatitis.

Description

PBMCs from 10 patients were cultured for 48 hours with no supplementation or supplemented with N-acetylcysteine (NAC), selenium or zinc sulfate. Genome-wide gene expression was assessed using the Affymetrix Clariom S Human HT microarray.

Each patient sample was analysed under all four culture conditions, allowing paired comparison of supplemented and unsupplemented PBMCs. Expression data were processed using robust multi-array average (RMA) normalisation in R/Bioconductor and differential expression was assessed using limma with patient identity included as a blocking factor.
