RNA-seq data of undifferentiated tonsil-derived mesenchymal stem cells from different donors
Listed in NCBI GEO
Donor-dependent variation in the differentiation potential of mesenchymal stem cells is an important consideration for their application in regenerative medicine.
Description
To investigate the molecular mechanisms underlying donor-dependent differences in skeletal myogenic differentiation, we performed RNA sequencing and compared the transcriptomic profiles of tonsil-derived mesenchymal stem cells (TMSCs) between donors with high (Myo-High) and low (Myo-Low) myogenic differentiation potential.
Donors #1 and #2 were classified as the Myo-High group, whereas Donors #3 and #4 were classified as the Myo-Low group based on their myogenic differentiation capacity. Differentially expressed gene (DEG) analysis identified a total of 462 genes with a fold change ≥ 2 and p < 0.05. Among these, 201 genes were significantly upregulated and 261 genes were downregulated in the Myo-High group compared with the Myo-Low group.

Further transcriptomic analysis identified POSTN as a candidate gene associated with high myogenic potential. Subsequent analyses demonstrated that POSTN was associated with autophagy regulation through the p-GSK3β (Ser9)–BCL2 axis. These findings suggest that donor-dependent differences in intrinsic autophagic capacity contribute to the myogenic differentiation potential of TMSCs and identify POSTN as a potential biomarker for selecting donors with superior myogenic potential.
